Abstract
    For the western part of the Dutch Wadden Sea primary production of phytoplankton and microphyobenthos as well as the input of organic matter from the North Sea are being investigated, but data on primary production of macrophytes are still lacking. In order to fill in this gap this study was started. Special attention was paid to finding the best method for measuring in situ primary production. The 14C-method proved to be superior to the O2-method, because of the frequent appearance of air bubbles in the oxygen bottles caused by warming up of the bottles during incubation on the tidal flat. Dry combustion and subsequent collection of the 14CO2 in a mixture of ethanolamine and 2-methoxyethanol gave the best results if the algae had to be prepared for liquid-scintillation-counting. This counting was carried out in a mixture of toluene (containing P.P.O. and P.O.P.O.P.) and 2-methoxyethanol and gave efficiency-values up to 80%. Comparison of the 14C-method with tbe O2-method (Gilson Respirometer) gave comparable results. Excretion of soluble organic compounds was found to be low (less than 3% of the primary production rates) to absent in all the experiments and measurements described.
