Abstract
My purpose here is to show how I have arrived at a new model of the voltage-gated sodium channel that for the first time successfully reconciles almost all the data obtained in recent years from a variety of experimental approaches. I have achieved this by exploiting to the full the superlative temporal resolution that can be achieved in recording the ionic and gating currents in the squid giant axon, and by relating the results to Numa's structural evidence (Noda et al. 1984) that each channel is controlled by four voltage-sensors arranged in parallel. For an updated account of these studies and a further revised model of the sodium channel, the reader is referred to a review by Keynes (1994).
