Abstract
Mutagenesis studies have been classically used to study development. However, mutagenesis has not typically been used to study physiological processes in adults. We have used a chemical mutagen, ENU, in combination with an F2 crossing scheme to recover mutations affecting ovarian and testicular maturation and gametogenesis in the adult zebrafish. To date we have recovered mutations which affect both spermatogenesis and oogenesis, cause proliferative stromal growth and/or hypertrophied follicle walls, induce large numbers of resorbing oocytes, and cause hypertrophied post-ovulatory follicles. We have confirmed five of these mutations to be present in the germline, and are currently examining 10 additional mutations for germline transmission. Mutations that are verified to be present in the germline will be genetically mapped using SSLP markers and further characterized cytologically and molecularly.
