Abstract
The action of recombinant human Activin A (rhActivin A) and a steroid/GnRH-free salmon testis extract on the production and release of gonadotropins in male coho at early stages of maturation has been investigated using a static dispersed pituitary cell culture system. rhActivin A produced a relatively weak increase (36%) in media FSH (LH remaining non-detectable) whilst, in contrast, the testis extract produced a large (157%) increase in FSH and LH rose to detectable levels. A multiplex RNase protection assay indicated that the testis extract and sGnRH, but not rhActivin A, significantly elevated alpha 1, alpha 2, FSH beta (LH beta non-detectable) subunit mRNA in pituitary cells incubated with these factors for 72 hours.
